Psychiatric genetics
The working group "psychiatric genetics" investigates the biological basis of neuropsychiatric disease. The focus of the working group is on the investigation of the genetic causes of mood disorders (bipolar disorder, depression), and anxiety disorders (panic disorder, social phobia). In the long term, improved understanding of the molecular basis of neuropsychiatric disease will lead to the development of new drugs and to targeted, individual therapeutic approaches.
Clinical features and importance of mood disorders and anxiety disorders
The main symptom of a mood disorder is a change in mood or affect. A distinction is made between unipolar depressive- and bipolar disorders. Among others, patients with depression present with depressed mood, a loss of interest, and a lack of drive. Patients with a bipolar disorder experience both depressive and manic episodes.
Clinically, panic disorder and social phobia belong to the anxiety disorder spectrum. Panic disorder patients experience sudden attacks of anxiety, so-called panic attacks, which occur for no apparent reason. The main symptom of social phobia is the fear of behaving in a way that will be embarrassing or humiliating while in the company of other people and of being negatively appraised.
Affective disorders and anxiety disorders are common, and rank among the main causes worldwide of a severe impairment in the activities of daily living. Improved understanding of the underlying biological mechanisms is therefore warranted.
Genetic basis
Affective disorders and anxiety disorders are multifactorial diseases. This means that both genetic factors and environmental factors contribute to their development.
The results of twin and family studies have long shown that genetic factors make a significant contribution to the development of mental illness. Estimated heredity (heritability) is around 60-85% for bipolar disorder, and around 40% for depression, panic disorder, and social phobia.
Recent molecular genetic investigations (so-called genome-wide association studies) of mood disorders have identified a number of risk genes, and our working group made a substantial contribution to these findings (including Stahl *, Breen *, Forstner *, McQuillin *, Ripke * et al., 2019; Wray et al., 2018). In the case of panic disorder and social phobia, the responsible genetic risk factors remain largely unknown. However, based on results generated for other mental illnesses, we can assume that future investigations of larger case-control collectives will identify genome wide significant genomic regions for these disorders.
Research concept of the working group "psychiatric genetics"
The focus of our working group is the identification of genetic risk factors for mood disorders and anxiety disorders. For this purpose, we apply high throughput molecular genetic technologies, such as SNP or methylation arrays, as well as diverse sequencing technologies (Sanger sequencing, next-generation sequencing, third-generation sequencing). Statistical and bioinformatic evaluation of the collected data involve single marker analyses, as well as innovative methods that take into account the cumulative contribution of numerous genetic markers (so-called polygenic risk scores), or the accumulation of association signals in specific metabolic pathways (so-called pathway analyses).
Our working group is very well networked both nationally (e.g., DFG research group 2107, for2107.de; Social Phobia Research, www.socialphobiaresearch.de), and internationally (e.g., Psychiatric Genomics Consortium, www.med.unc.edu/pgc/), and has access to large patient and control cohorts.
The systematic elucidation of the biological causes of mental illness is an important starting point for the development of new therapeutic strategies. In addition, genetic findings could contribute to the establishment of new diagnostic concepts.
